吉西他濱加順鉑(GP)化療是鼻咽癌(NPC)的一種標準治療方式。然而，治療的機制尚不清楚。最近，中山大學(xué)科研團隊在期刊《Nature Medicine》（IF=82.9）上發(fā)表題為“The tumor immune microenvironment of nasopharyngeal carcinoma after gemcitabine plus cisplatin treatment.”的文章，使用單細胞RNA測序，對首次治療和GP治療后的NPC樣本(n = 15對)的T細胞和B細胞受體測序匹配，結(jié)果顯示GP化療會激活先天樣B細胞(ILB)主導(dǎo)的抗腫瘤免疫反應(yīng)?；熣T發(fā)的DNA片段激活了STING, 增加干擾素依賴途徑在癌細胞中表達MHC Class I，并同時誘導(dǎo) ILB通過Toll樣受體9信號通路。對 germinal centers無效的化療，ILB通過ICOSL–ICOS軸進一步擴大了follicular helper和helper type 1 T 細胞隨后增強三級淋巴器官樣結(jié)構(gòu)中的細胞毒性T細胞。ILB頻率是與3期臨床試驗中的接受GP化療的NPC患者(NCT01872962，n = 139)總體無病生存率呈正相關(guān)。ILB頻率可以作為GP和免疫療法聯(lián)合治療(n = 380)以下治療的NPC患者中作為有利結(jié)果的預(yù)測因子。該研究提供了GP化療后腫瘤免疫微環(huán)境的高分辨率地圖并發(fā)現(xiàn)B細胞為中心的抗腫瘤作用。研究還確定并驗證了在NPC以全科醫(yī)生為基礎(chǔ)的治療中ILB作為一個潛在的生物標志物可以改善病人管理。
本文中使用了欣博盛Human IFN-β ELISA kit檢測了細胞培養(yǎng)上清中該蛋白的含量。
