微環(huán)境中的代謝變化顯著調(diào)節(jié)腫瘤免疫敏感性，但其潛在機制仍不清楚。最近，清華大學生命科學學院團隊在期刊《Cell Metabolism》（IF=31.373）上發(fā)表題為“Cancer-cell-derived fumarate suppresses the anti-tumor capacity of CD8+ T cells in the tumor microenvironment.”的文章，報道了富馬酸水合酶(FH)缺失的腫瘤表現(xiàn)出對功能性CD8+ T細胞活化、擴增和功效的抑制，并具有增強的惡性增殖能力。推斷腫瘤細胞中的FH缺失會在腫瘤間質(zhì)液中積累延胡索酸，增加的延胡索酸可直接在C96和C102處琥珀酸化ZAP70，并減弱其在浸潤CD8+ T細胞中的活性，導致CD8+ T細胞活化受到抑制，并在體外和體內(nèi)產(chǎn)生抗腫瘤免疫反應(yīng)。此外，通過增加FH表達來消除延胡索酸鹽可顯著增強抗CD19 CAR T細胞的抗腫瘤功效。該研究證明了富馬酸在控制TCR信號中的作用，并表明腫瘤微環(huán)境(TME)中的富馬酸積累是CD8+ T細胞抗腫瘤功能的代謝障礙。該研提出了延胡索酸的減少可能是腫瘤免疫治療的一個重要策略。
本文中使用了欣博盛Human Granzyme B ELISA kit檢測了細胞培養(yǎng)上清中這個細胞因子的含量。
