近期，華東理工大學(xué)劉昌勝院士和王靖教授團(tuán)隊(duì)在Bioactive Materials（IF=18.0）上發(fā)表題為“Immunoregulative coating for scarless healing in anterior cruciate ligament reconstruction（免疫調(diào)節(jié)涂層在前交叉韌帶重建無瘢痕愈合中的應(yīng)用）”的文章，在該項(xiàng)研究中，研究團(tuán)隊(duì)開發(fā)了一種多層免疫調(diào)節(jié)水凝膠涂層，用于無瘢痕的PET（Polyethylene terephthalate）-骨整合。這種多層炎癥調(diào)節(jié)水凝膠涂層的應(yīng)用不僅可以去除瘢痕組織屏障，還可以通過增強(qiáng)血管生成和成骨來改善移植物-骨骼整合。此外，它避免了外源性生長(zhǎng)因子的過度使用和潛在的并發(fā)癥，提供了一種更方便可行的治療策略。
SCS@PGH/PET水凝膠涂層在研究中顯示了卓越的特性，包括為細(xì)胞滲透而優(yōu)化的多孔結(jié)構(gòu)，以及增加的對(duì)細(xì)胞粘附和增殖至關(guān)重要的潤(rùn)滑性。再結(jié)合硫酸化多糖和生物活性分子已被證明在調(diào)節(jié)巨噬細(xì)胞(Mφs )從促炎性M1狀態(tài)到再生性M2狀態(tài)的極化中至關(guān)重要（調(diào)節(jié)了如TGF-β這樣的細(xì)胞因子），而這種轉(zhuǎn)變對(duì)于減少瘢痕形成、促進(jìn)愈合環(huán)境和加速血管生成至關(guān)重要。通過體外和體內(nèi)研究，團(tuán)隊(duì)確認(rèn)了水凝膠對(duì)組織修復(fù)和再生的顯著影響?？傮w而言，SCS@PGH/PET水凝膠涂層的發(fā)現(xiàn)為再生醫(yī)學(xué)中合成韌帶的設(shè)計(jì)和改進(jìn)提供了堅(jiān)實(shí)的科學(xué)基礎(chǔ)。
