近期，上海交通大學(xué)藥學(xué)院沈琦團(tuán)隊在Advanced Functional Materials（IF=18.5）上發(fā)表題為“Self-Adjuvanting Bacteria Hydrogel for SHP1 Checkpoint Inhibition in Tumor-Draining Lymph Nodes to Enhance Cancer Immunotherapy.（自佐劑細(xì)菌水凝膠抑制腫瘤引流淋巴結(jié)SHP1檢查點增強(qiáng)癌癥免疫治療）”的研究。盡管免疫療法最近取得了進(jìn)展，但其效果仍受到缺乏免疫協(xié)調(diào)的限制。特別是腫瘤引流淋巴結(jié)(TDLNs)與腫瘤之間的相互作用經(jīng)常被忽視。在該項研究中，團(tuán)隊開發(fā)了一種能夠激發(fā)強(qiáng)大且持久免疫反應(yīng)的自佐劑水凝膠。該AAM水凝膠系統(tǒng)通過抑制腫瘤引流淋巴結(jié)中的SHP1免疫檢查點，并增強(qiáng)腫瘤細(xì)胞因子效應(yīng)，協(xié)同誘導(dǎo)抗腫瘤免疫反應(yīng)，為安全而有效的抗腫瘤免疫治療提供了一種創(chuàng)新策略。

表達(dá)IL-15的工程化的阿拉伯糖反應(yīng)細(xì)菌(ARB)和負(fù)載維生素E的甘露糖修飾的中空介孔普魯士藍(lán)納米顆粒(NP)(Man/HMPB(VE),MHV)與阿拉伯糖水凝膠(AraGel)混合，形成AraGel@ARB/MHV(AAM)系統(tǒng)。通過MHV介導(dǎo)的溫和光熱療法，免疫原性細(xì)胞死亡(ICD)觸發(fā)腫瘤相關(guān)抗原的釋放。隨后，Man修飾的NP靶向TDLN并釋放VE，VE會抑制樹突狀細(xì)胞中含檢查點Src同源區(qū)2結(jié)構(gòu)域的磷酸酶-1(SHP1)，從而增強(qiáng)抗原呈遞和T細(xì)胞的激活。同時，AraGel的降解會誘導(dǎo)ARB(IL-15)表達(dá)IL-15，這使得ARB能夠以自佐劑的方式作為增強(qiáng)佐劑，與ICD和TDLN重編程協(xié)同作用，促進(jìn)細(xì)胞毒性T淋巴細(xì)胞的激活。該水凝膠系統(tǒng)通過以協(xié)同的方式激發(fā)持久且強(qiáng)大的免疫治療，有效地抑制了腫瘤的生長?？偠灾?，自佐劑水凝膠在癌癥免疫治療方面具有巨大潛力。
