皮膚惡性黑色素瘤,來源于產(chǎn)生色素的黑素細胞,容易擴散到淋巴或血管中,嚴重威脅黑色素瘤患者的生存質(zhì)量和存活率。常規(guī)的化療和放療治療不足以克服晚期黑色素瘤的頑固性。研究表明PD-1和PD-L1的相互作用通過mTOR過度激活加劇周圍侵襲,從而推動腫瘤發(fā)生、血管生成和放射抵抗。研究探索阻斷黑色素瘤內(nèi)在生長信號,包括同時阻斷PD-L1和mTOR信號,配合放療,可以提供一個強有力的措施來減輕腫瘤的負擔。最近,澳門大學的研究團隊在期刊《Bioactive Materials》(IF=16.874)上發(fā)表題為“A platinum@ polymer-catechol nanobraker enables radio-immunotherapy for crippling melanoma tumorigenesis, angiogenesisstance., and radioresistance.”的文章,設計了一種三叉鉑@聚合物-兒茶酚納米braker來遞送mTOR抑制劑TAK228和抗PD-L1抗體(aPD-L1 ),以阻止黑色素瘤中PD-1驅(qū)動的侵襲并最大限度地根除黑色素瘤。aPD-L1協(xié)同TAK228通過阻斷黑色素瘤-PD-L1配體和黑色素瘤-PD-1受體驅(qū)動的mTOR信號傳導抑制PD-1:與PD-L1致瘤性相互作用;mTOR下游原生細胞內(nèi)源MYC和原生血管缺氧誘導因子1-α的相應下調(diào)分別有助于預防腫瘤發(fā)生和血管生成。此外,high-Z metal platinum增敏TAK228增強的放射療法賦予納米braker顯著的殺腫瘤功效。三叉鉑@聚合物-兒茶酚納米braker能有效地抑制了黑色素瘤腫瘤發(fā)生和血管生成,同時增強了放射治療功效。這種巧妙的策略可能會給臨床黑色素瘤患者帶來好處。


本文中使用了欣博盛Mouse TNF-α, IL-6, IL-12p70 ELISA kits檢測了血清樣本中這些細胞因子的含量。
