缺血性心臟病是由于冠狀動(dòng)脈疾病引起的心肌變性、壞死和纖維化,并導(dǎo)致嚴(yán)重左室功能障礙的一種疾病,是導(dǎo)致心力衰竭常見(jiàn)的原因,通過(guò)成功開(kāi)通閉塞的冠狀動(dòng)脈治療心肌梗死的可顯著降低死亡率,因此,學(xué)者針對(duì)左心室病理性重塑進(jìn)行了大量研究。心室重塑與心臟功能和結(jié)構(gòu)的變化有關(guān),臨床和實(shí)驗(yàn)研究都表明炎癥反應(yīng)參與心肌梗死后心室重塑,清除梗死區(qū)域死亡細(xì)胞和細(xì)胞外基質(zhì)碎片,然而,過(guò)度活躍的炎癥信號(hào)會(huì)加劇心肌損傷并導(dǎo)致不良的心室重塑。近日,華中科技大學(xué)同濟(jì)醫(yī)學(xué)院附屬協(xié)和醫(yī)院心血管內(nèi)科夏霓和程翔教授團(tuán)隊(duì)在期刊《Basic research in cardiology》(IF: 17.165)上發(fā)表了題為“Regulatory B cells improve ventricular remodeling after myocardial infarction by modulating monocyte migration”的文章,旨在確定調(diào)節(jié)性B細(xì)胞(Bregs)對(duì)心室重構(gòu)的影響,并探討其相關(guān)機(jī)制。本研究發(fā)現(xiàn)過(guò)繼轉(zhuǎn)移Bregs可改善小鼠心肌梗死模型中的心室重構(gòu),如改善心臟功能、減少瘢痕大小和減輕間質(zhì)纖維化,但不改變存活率。從機(jī)制上講,Bregs降低了單核細(xì)胞中C-C基序趨化因子受體2(CCR2)的表達(dá),這抑制了促炎性單核細(xì)胞從外周血向心臟募集和骨髓動(dòng)員。用白細(xì)胞介素10(IL-10)抗體治療可消除Breg介導(dǎo)的心肌梗死保護(hù)作用。最后,IL-10中和作用改變了Bregs對(duì)單核細(xì)胞遷移和CCR2表達(dá)的影響。目前的研究表明,通過(guò)減少CCR2介導(dǎo)的單核細(xì)胞募集和動(dòng)員,Bregs在限制心肌梗死后心室重構(gòu)方面具有治療價(jià)值。


本文中使用了欣博盛小鼠IL-10和TGF-β1 ELISA試劑盒,檢測(cè)了小鼠脾臟組織中IL-10的表達(dá)量以及小鼠脾臟調(diào)節(jié)性B細(xì)胞和對(duì)照B細(xì)胞中IL-10和TGF-β1的表達(dá)情況。
