近期,武漢大學(xué)黃衛(wèi)華團(tuán)隊、廈門大學(xué)Christian Amatore團(tuán)隊合作在《Nature Nanotechnology》(IF=38.8)上發(fā)表題為“Nanosensor detection of reactive oxygen and nitrogen species leakage in frustrated phagocytosis of nanofibres”(納米電極監(jiān)測巨噬細(xì)胞受挫性吞噬納米纖維過程中ROS/RNS泄漏)的研究,報告了一種電化學(xué)納米傳感器,用于監(jiān)測和定量表征單個巨噬細(xì)胞對玻璃納米纖維的吞噬作用受阻期間活性物質(zhì)釋放的通量和動態(tài)。
機(jī)體吸入環(huán)境暴露的高長徑比惰性納米纖維(石棉、玻璃纖維、碳納米管等)后,肺泡巨噬細(xì)胞會迅速識別這些異物,并試圖將其吞噬。由于纖維狀材料的高長徑比和難于分解的特性,巨噬細(xì)胞難以將其快速清除,從而形成受挫性吞噬作用(frustrated phagocytosis)。在此過程中,巨噬細(xì)胞內(nèi)的NADPH氧化酶(NOX)和誘導(dǎo)型一氧化氮合酶(iNOS)會持續(xù)產(chǎn)生活性氧和活性氮(ROS/RNS)消化纖維。由于吞噬腔未封閉,大量產(chǎn)生的ROS/RNS可能會從吞噬口泄漏到胞外,對周圍肺組織細(xì)胞造成損傷并引發(fā)疾病。在過去幾十年里,研究人員采用ROS/RNS染色以及活體實驗對碳納米管和石棉等納米纖維材料的研究均支持這一假設(shè)。然而,因缺乏合適的傳感器,該過程中各類活性物質(zhì)的化學(xué)組成及其時空動態(tài)變化特征仍然未知。
針對受挫性吞噬作用假說,研究團(tuán)隊將鉑黑納米線電化學(xué)傳感器(SiC@PtNWS)定位在巨噬細(xì)胞吞噬口處,從而實現(xiàn)對受挫性吞玻璃納米纖維的過程中4種主要ROS/RNS(ONOO-,H2O2,NO和NO2-)泄漏通量的實時定量監(jiān)測。結(jié)果表明,在吞噬各階段,ROS/RNS釋放存在明顯時間差異。相較于細(xì)胞頂部,吞噬口處存在大量ROS/RNS泄漏,且隨吞噬進(jìn)行持續(xù)增加直到吞噬口封閉泄漏。NOX和iNOS參與動態(tài)調(diào)控ROS/RNS的物質(zhì)組成。相關(guān)數(shù)據(jù)定量展示了單個巨噬細(xì)胞受挫性吞噬作用玻璃納米纖維時ROS/RNS產(chǎn)生的時空特征,從而描繪出巨噬細(xì)胞應(yīng)對惰性高長徑納米材料的動態(tài)免疫調(diào)節(jié)過程。體外共培養(yǎng)和活體實驗進(jìn)一步表明,持續(xù)且大量的ROS/RNS泄漏會導(dǎo)致外周細(xì)胞嚴(yán)重受損,最終轉(zhuǎn)化為慢性炎癥和肺損傷。
在該項研究中,使用了欣博盛生物(NeoBioscience Technology Co, Ltd)的QuantiCyto? Mouse IL-1α ELISA kit(小鼠白細(xì)胞介素-1α),用于培養(yǎng)上清液中該細(xì)胞因子含量的測定。
