動脈粥樣硬化是許多心血管疾病中常見的病理現(xiàn)象。盡管目前的療法(包括他汀類藥物和絲氨酸蛋白酶PCSK9抑制劑)可以有效地將低密度脂蛋白(LDL)膽固醇水平降低到臨床指南推薦的水平,但心血管不良事件仍然頻繁發(fā)生。脂蛋白在動脈壁的內(nèi)皮下滯留觸發(fā)了巨噬細胞的多種炎癥事件,是動脈粥樣硬化病理進展的主要原因。因此,調(diào)節(jié)炎癥是預防和治療動脈粥樣硬化及其并發(fā)癥的一種途徑。不幸的是,在控制斑塊炎癥的特異性和有效性方面的挑戰(zhàn)阻礙了動脈粥樣硬化治療的進展。最近,上海交通大學研究團隊在期刊《ACS nano》(IF=17.1)上發(fā)表題為“Modulating Plaque Inflammation via Targeted mRNA Nanoparticles for the Treatment of Atherosclerosis”的文章,報道了一種NP介導的mRNA治療方法,以動脈粥樣硬化病變巨噬細胞為靶點,調(diào)節(jié)晚期動脈粥樣硬化病變中的炎癥,從而治療動脈粥樣硬化。證明了含有IL-10 mRNA的靶向NPs與M2樣巨噬細胞共定位,并在靜脈注射給Western diet(WD)喂養(yǎng)的Ldlr小鼠后誘導動脈粥樣硬化斑塊中IL-10的產(chǎn)生。此外,氧化應激和巨噬細胞凋亡減少證明損傷顯示出炎癥反應顯著減輕,導致脂質(zhì)沉積減少,壞死面積減少,纖維帽厚度增加。這些結果表明,在晚期動脈粥樣硬化的臨床前模型中,將mRNA治療劑成功遞送至富含巨噬細胞的斑塊,表明這種靶向NP炎癥方法具有轉化為廣泛臨床應用的巨大潛力。
本文中使用了欣博盛Mouse IL-10, IL-1β和 IL-6 ELISA kits檢測了主動脈組織和血清樣本中目標蛋白的含量。
