特發(fā)性肺纖維化(IPF, Idiopathic pulmonary fibrosis)是一種慢性進行性間質(zhì)性肺病,最終會導(dǎo)致肺功能永久惡化并死亡。白細胞介素-11 (IL-11)是一種促纖維化細胞因子,對成纖維細胞分化為分泌膠原、肌動蛋白α2、平滑肌陽性(ACTA2+)肌成纖維細胞至關(guān)重要,是特發(fā)性肺纖維化(IPF)發(fā)病機制的驅(qū)動過程。最近,上海交通大學(xué)藥學(xué)院在期刊《Science Advances》(IF=14.957)上發(fā)表題為“Inhaled siRNA nanoparticles targeting IL11 inhibit lung fibrosis and improve pulmonary function post-bleomycin challenge.”的文章,開發(fā)了一種可吸入和可粘液滲透的納米顆粒(NP)系統(tǒng),該系統(tǒng)結(jié)合了抗IL11的siRNA(siIL11 @ PPGC NPs ),并研究了其治療IPF的治療潛力。NP是通過可生物降解的PLGA-PEG二嵌段共聚物和自創(chuàng)的陽離子類脂分子G0-C14的自組裝來配制的,以實現(xiàn)siIL11的有效跨粘膜遞送。非侵入性霧化吸入通過抑制ERK和SMAD2來阻止成纖維細胞分化和減少ECM沉積。此外,在博萊霉素誘導(dǎo)的肺纖維化的小鼠模型中,siIL11@PPGC NPs顯著減少了纖維化的發(fā)展并改善了肺功能,而沒有誘導(dǎo)全身毒性。這項研究為siRNA治療劑的局部吸入給藥提供了一個通用的納米顆粒平臺,并在許多呼吸系統(tǒng)疾病(包括IPF)的治療中顯示出有希望的臨床潛力。
