卵巢癌是女性生殖器官最常見(jiàn)的惡性腫瘤之一，是婦科惡性腫瘤的主要死亡原因，晚期5年生存率不到42%。近年來(lái)，基于納米顆粒的治療平臺(tái)出現(xiàn)，給卵巢癌患者帶來(lái)了新的希望。然而，未經(jīng)修飾的常規(guī)納米藥物可被網(wǎng)狀內(nèi)皮系統(tǒng)快速清除，導(dǎo)致有限的治療效果。目前研究的一種細(xì)胞膜涂層策略來(lái)賦予納米粒子仿生功能，涂層納米粒子保留了核心成分的物理化學(xué)特性，同時(shí)具有使人工材料擁有難以復(fù)制的復(fù)雜細(xì)胞膜功能。最近，武漢大學(xué)中南醫(yī)院張蔚教授團(tuán)隊(duì)在期刊《ACS nano》（IF:15.881）上發(fā)表了題為”Cancer-Erythrocyte Hybrid Membrane-Camouflaged Magnetic Nanoparticles with Enhanced Photothermal-Immunotherapy for Ovarian Cancer”的文章，將小鼠來(lái)源的ID8卵巢癌細(xì)胞膜與紅細(xì)胞(RBC)膜融合以產(chǎn)生混合仿生涂層(IRM)，并制備了混合IRM偽裝的吲哚菁綠(ICG)負(fù)載磁性納米粒子(Fe3O4-ICG@IRM)用于卵巢癌的聯(lián)合治療。Fe3O4-ICG@IRM同時(shí)保留了ID8和RBC細(xì)胞膜蛋白，并在體外和體內(nèi)表現(xiàn)出對(duì)ID8細(xì)胞的高度特異性自我識(shí)別以及延長(zhǎng)的血液循環(huán)壽命。在雙側(cè)腫瘤模型中，IRM包被的納米顆粒也激活了特異性免疫，殺死了同源的ID8腫瘤細(xì)胞，但對(duì)B16-F10腫瘤細(xì)胞沒(méi)有影響。此外，F(xiàn)e3O4-ICG@IRM顯示了協(xié)同光熱治療，通過(guò)光熱誘導(dǎo)的腫瘤壞死導(dǎo)致全細(xì)胞腫瘤抗原的釋放，這通過(guò)激活CD8+細(xì)胞毒性T細(xì)胞和減少調(diào)節(jié)性Foxp3+ T細(xì)胞進(jìn)一步增強(qiáng)了對(duì)原發(fā)性腫瘤和轉(zhuǎn)移性腫瘤的抗腫瘤免疫治療。
本文中使用了欣博盛Mouse IL-1β, IL-6, TNF-α和IFN-γ檢測(cè)了細(xì)胞上清液細(xì)胞因子的表達(dá)量。
