多年來,大量研究證明免疫細(xì)胞和腫瘤細(xì)胞產(chǎn)生的內(nèi)源性IFN-I在調(diào)控腫瘤生長(zhǎng)和抗腫瘤治療中有重要作用。IFN-I在血液惡性腫瘤和實(shí)體瘤中都有良好的臨床療效。目前,臨床上可以通過注射重組人IFN提高其瘤內(nèi)水平,但其腫瘤靶向性差、療效低,且易產(chǎn)生明顯的全身免疫毒性。因此,如何選擇性提高瘤內(nèi)IFN等免疫分子水平,同時(shí)克服其誘導(dǎo)的免疫耐受,是腫瘤免疫治療領(lǐng)域亟待解決的重要科學(xué)問題。近日,中國(guó)科學(xué)院上海藥物研究所李亞平課題組在國(guó)際頂級(jí)期刊《Nature Nanotechnology》上在線發(fā)表了題為”T lymphocyte membrane-decorated epigenetic nanoinducer of interferons for cancer immunotherapy”的文章,展示了工程化T淋巴細(xì)胞膜修飾干擾素(IFN)表觀遺傳納米誘導(dǎo)劑改善腫瘤免疫治療的最新研究成果。該團(tuán)隊(duì)創(chuàng)造性地設(shè)計(jì)構(gòu)建了一種仿生納米囊泡,將藥物ORY-1001包載到PD1高表達(dá)的T細(xì)胞膜囊中,構(gòu)建了一種新型免疫調(diào)節(jié)劑(簡(jiǎn)稱OPEN)。OPEN能特異性靶向腫瘤,高效誘導(dǎo)瘤內(nèi)IFN分泌,上調(diào)腫瘤細(xì)胞PDL1和主要組織相容性復(fù)合體-I(MHC-I)等的表達(dá)。該研究開拓了精準(zhǔn)遞送+智能釋藥一體化技術(shù)調(diào)控表觀遺傳、克服免疫耐受、改善腫瘤免疫治療的新方向,為提高包括IFN在內(nèi)的兼具抗癌活性和促進(jìn)免疫逃逸的活性分子療效以及降低其免疫副作用提供了新思路。


本文中使用了欣博盛Mouse IFN-α和IFN-β ELISA試劑盒檢測(cè)了細(xì)胞培養(yǎng)上清和腫瘤組織勻漿樣本中的IFN-α和IFN-β濃度,使用Mouse PD1 ELISA試劑盒檢測(cè)了OPEN上PD-1蛋白的濃度。
