炎癥性腸病(IBDs)是一種全球性疾病,發(fā)病率在發(fā)展中國家呈上升趨勢,該病常反復(fù)發(fā)作,嚴重影響患者的正常生活。生物鐘系統(tǒng)是生物體內(nèi)生的“振蕩器”,控制著多種生理和行為進程,生物鐘與炎癥性疾病關(guān)系密切,生物鐘還可能是炎癥性疾病癥狀產(chǎn)生晝夜差異的重要因素。盡管晝夜節(jié)律如何影響炎癥越來越明了,但炎癥調(diào)節(jié)晝夜節(jié)律的分子機制仍然難以捉摸。LncRNA(長非編碼RNA)是一類非編碼RNA,可通過核苷酸堿基配對或RNA折疊產(chǎn)生的結(jié)構(gòu)域的形成,與DNA,其他RNA和蛋白質(zhì)發(fā)生相互作用,從而調(diào)節(jié)基因轉(zhuǎn)錄。一些lncRNA與免疫反應(yīng)有關(guān),在炎癥反應(yīng)(如內(nèi)毒素血癥和腎炎)的發(fā)生中發(fā)揮作用。另外,lncRNA對生物鐘有潛在的影響,還可能受生物鐘的控制。然而,lncRNA在結(jié)腸炎與晝夜節(jié)律的交互作用中的作用在很大程度上是未知的。


近期,暨南大學吳寶劍教授團隊在Science Advances上發(fā)表了題為” An NF-κB-driven lncRNA orchestrates colitis and circadian clock ”的研究論文,該研究發(fā)現(xiàn)NF-κB驅(qū)動的一種周期性表達的lncRNA——Lnc-UC,能通過表觀遺傳機制調(diào)控有抗炎作用的節(jié)律基因Rev-erbα的表達,從而將晝夜節(jié)律和結(jié)腸炎聯(lián)系起來。靶向Lnc-UC可能是干預(yù)炎癥與生物鐘之間的互相作用,并控制結(jié)腸炎的新策略。


本文中使用了欣博盛Mouse IL-1β and IL-18 ELISA試劑盒,檢測了小鼠骨髓來源的巨噬細胞培養(yǎng)基中IL-1β和IL-18的濃度。
